【摘要】 目的 探讨葛根素对大鼠脑缺血后脑促红细胞生成素（epo）表达的影响。方法 应用线栓法制备大鼠大脑中动脉缺血2 h再灌注48 h损伤模型；分别于缺血2 h再灌注即刻、再灌注24 h给予腹腔注射葛根素400 mg/kg（体重），对照组腹腔注射等剂量生理盐水，每组12只大鼠。大鼠苏醒后30 min及处死前分别进行神经功能评分。48 h处死大鼠，每组6只石蜡切片用he染色检测腔梗死体积，免疫组织化学检测葛根素对缺血脑组织epo表达的影响；每组6只elisa法检测epo活性。结果 葛根素400 mg/kg（体重）腹腔注射组的脑梗死体积明显小于对照组，葛根素治疗也可减少神经功能损伤，神经功能评分均明显好于对照组。免疫组织化学结果显示：葛根素组大鼠脑缺血侧皮层区及基底区epo阳性细胞数明显高于对照组。结论 葛根素对大鼠脑缺血再灌注损伤具有保护作用。其保护作用可能与提高epo的表达有关。

【abstract】 objective to test whether chinese medicine puerarin has effects in erythropoietin (epo) expression in rats following middle cerebral artery occlusion (mcao). methods ischemiareperfusion injury (i/r injury) was induced by ligated mcao with 2 h ischemia and 48 h reperfusion. 400 mg/kg puerarin or saline (control) were administered intraperitoneally at two points: immediately and 48 h after reperfusion. the rats were sacrificed 48 h after reperfusion. infarct volume was measured through triphenyl tetrazolium chloride (ttc) staining. the epo expressions were determined by immunohistochemistry. functional neurological outcome was determined at 30 min and 48 h after the rats awake from the surgery. results compared to control group, the infarct volume of 400 mg/kg puerarin group decreased obviously. the puerarintreated group had better functional neurological outcome compared to the controls. the epo expressions were increased obviously in puerarintreated group. conclusions puerarin has neuroprotective effects on rats with i/r injury, one of the underlying mechanism may through increasing epo expressions during i/r injury.......
